RIPK3 (Receptor Interacting Serine/Threonine Kinase 3) is a protein coding gene located on human chromosome 14q12. RIPK3 is also known as RIP3. The human RIPK3 gene encodes a 56887 Da protein containing 518 amino acids. The RIPK3 protein is broadly expressed in small intestine, duodenum and other tissues. Among its related pathways are Activated TLR4 signalling and IKK complex recruitment mediated by RIP1. RIPK3 is related to identical protein binding and protein kinase activity. RIPK2 is an important paralog of RIPK3 gene. RIPK3 is associated with some diseases, including Caspase 8 Deficiency and Zika Virus Congenital Syndrome.

    Necroptosis is a form of programmed necrotic cell death that is rapidly emerging as an important pathophysiological pathway in numerous disease states. Necroptosis is dependent on receptor-interacting protein kinase 3 (RIPK3), a protein shown to play an important role in experimental models of critical illness. RIPK3 promotes kidney tubular injury via mitochondrial dysfunction. The FAO-dependent RIPK3 mediates pathogenesis of acute lung injury.
