PHF1 (PHD Finger Protein 1) is a protein coding gene located on human chromosome 6p21.32. PHF1 is also known as PCL1, PHF2, hPHF1, MTF2L2, and TDRD19C. The human PHF1 gene encodes a 62106 Da protein containing 567 amino acids. The PHF1 protein is ubiquitously expressed in ovary, testis and other tissues. Among its related pathways are Activated PKN1 stimulates transcription of AR (androgen receptor) regulated genes KLK2 and KLK3 and Gene Expression. PHF1 is related to DNA-binding transcription factor activity and methylated histone binding. MTF2 is an important paralog of PHF1 gene. PHF1 is associated with some diseases, including Endometrial Stromal Sarcoma and Endometrial Stromal Nodule.

    PHF1 の背景知識

    PHF1 gene rearrangements have been recently described in around 50% of ossifying fibromyxoid tumors (OFMT) including benign and malignant cases, with a small subset showing EP400-PHF1 fusions. PHF11 (plant homeodomain finger 11) encodes a previously unknown DDR factor involved in 5' end resection, ATR signaling, and HR. PHF11 was identified based on its association with deprotected telomeres and localized to sites of DNA damage in S phase. Depletion of PHF11 diminished the ATR signaling response to telomere dysfunction and genome-wide DNA damage, reduced end resection at sites of DNA damage, resulted in compromised HR and misrejoining of S-phase DSBs, and increased the sensitivity to DNA-damaging agents.
