PDZK1 (PDZ Domain Containing 1, also known as CAP70; CLAMP; PDZD1; NHERF3; NHERF-3), located on 1q21.1, is a Protein Coding gene. This gene encodes a PDZ domain-containing scaffolding protein. The gene produces a 57129 Da protein composed of 519 amino acids. PDZK1 is a multi-PDZ domain-containing adaptor protein that binds to the C terminus of the high-density lipoprotein receptor, scavenger receptor, class B, type I (SR-BI), and controls the posttranscriptional, tissue-specific expression of this lipoprotein receptor. Diseases such as Gout and Inflammatory Diarrhea are associated with PDZK1.

    PDZK1 の背景知識

    PDZK1 (Post-synaptic density protein/Drosophila disc-large protein/Zonula occludens protein containing 1) is an adaptor protein that binds to the HDL receptor, scavenger receptor class B type I. PDZK1 is a multi-PDZ domain-containing adaptor protein that binds to the C terminus of the high-density lipoprotein receptor, scavenger receptor, class B, type I (SR-BI), and controls the posttranscriptional, tissue-specific expression of this lipoprotein receptor. PDZK1 (NHERF3) interacts with membrane proteins whereby modulating their spatial arrangement, membrane stability, and function. PDZK1 is important for maintaining adequate steady-state levels of SR-BI in the liver but is not essential for cell surface expression or function of hepatic SR-BI. Leukocyte PDZK1 deficiency in high fat-diet fed LDL receptor knockout mice has been found to increase atherosclerotic necrotic core formation and apoptosis of cells within atherosclerotic plaques.
